A. Abdelnoor. Research Projects Bacterial DNA and plasmid DNA vaccines Genomic DNA was extracted from Salmonella typhimurium
and Pseudomonas aeruginosanosa.Virulence factor genes from these two
organisms were amplified by PCR, and amplicons-plasmid DNA recombinants were
prepared. The protective effects in mice of the gemonic DNA and recombinant
DNA against lethal challenge with the organisms are being tested. Abdelnoor,

rhinitis in Lebanon The purpose of this study is to identify causative
allergens in patients with allergic rhinitis in Lebanon and determine
possible correlations among serum allergen-specific antibody, polyclonal IgE,

the expression of E.coli 0157:H7 verocytotoxin gene The study deals with PCR-based detection, transcription
and possibly translation of verocytotoxin gene of E.coli 0157:H7, and
assessment of the role of certain agents on the blocking expression of the
protein. Matar, G. M. Supported by Lebanese National Council for Research Comparative analysis of virulence factors produced by P.aeruginosa
and severity of symptoms in patients with otitis externa A correlation between production of several virulence
factors by P.aeruginosa from patients having unilateral or bilateral
otitis externa and severity of symptoms was done. Data have shown that
detected virulence factors, including protease, elastase, fibrinolysin,
DNase, coagulase, lipase and lecithinase, were all produced by P.aeruginosa
isolates associated with severe symptoms while a few virulence factors,
mostly protease, elastase, and fibrinolysin, were produced by isolates
associated with mild to moderate symptoms. This indicates that severe
symptoms manifested by patients may be due to the overall production of the
virulence factors. Studies are underway to detect, quantitatively, the levels
of these virulence factors. Matar, Gh. M. Supported by URB Genotypic characteristics of E.coli 0157:H7 and
other pathogenic E.coli obtained from diarrheal stools The project deals with isolation of E.coli isolates
from diarrheal stools, and molecular detection of pathogenic isolates by
multiplex PCR. In addition, genotyping of detected pathogenic isolates within
each category will be carried out by Pulsed-Field Gel Electrophoresis to
determine possible strain variation among isolates. Matar. Gh. M. Completed or in progress at AUB Molecular characteristics of Haemophilus species
in head and neck infections Haemophilus isolates obtained from tonsillar sites
of patients undergoing tonsillectomy and adenoidectomy were identified
phenotypically and subtyped by RAPD analysis. Ton B gene encoding a virulence
factor the ton B protein was detected and analyzed by PCR-RFLP analysis in H.influenzae
and H.parainfluenzae. Ongoing work deals with detection of mRNA
transcripts levels in both Haemophilus species. Matar. G. M. The role of P.aeruginosa exotoxin A in relation
to severity of symptoms in patients with otitis externa The study dealt with PCR-based detection and transcription of P.aeruginosa exotoxin A gene in relation to severity of symptoms. Data have shown that exotoxin A gene is detected and transcribed in all tested isolates of P.aeruginosa, associated with severe and mild to moderate symptoms, in patients with otitis externa. Current studies are underway to detect transcription levels of P.aeruginosa exotoxin A gene and severity of symptoms in these patients. Matar, Gh. M. Supported by URB |
