Targeting of the ceramide pathway by viral proteins

Apoptosis is important in many aspects of homeostasis, cancer development and chemotherapy, and the immune response. One of the central regulators of the apoptotic response appears to be ceramide, a sphingolipid breakdown product. Treatment with an inducer of apoptosis, such as tumor necrosis factor a (TNFa), results in the elevation of endogenous ceramide that subsequently drives the cell towards apoptosis. Some viruses encode for proteins that disable this defensive response allowing the virus to produce more progeny before the eventual demise of the infected cell. We are studying several viral proteins that have been shown to have antiapoptotic activities and examining their effects on the ceramide pathway of apoptosis. Preliminary studies show that some viral proteins act upstream of ceramide generation by directly or indirectly inhibiting enzymes that result in ceramide generation. Other viral proteins appear to work downstream of ceramide by inhibiting effector molecules activated by ceramide. G. Dbeibo. (Supported by AUB MPP, LNCSR, URB.)
 
 

Modulation of the TNFa-induced ceramide pathway by E3 proteins of adenovirus

Adenoviruses contain a number of proteins that modulate the apoptotic response. Specifically, some adenoviral proteins encoded by the E3 region have been shown to inhibit TNFa-triggered apoptosis. In this project, we are investigating the effect of E3 proteins on the ceramide pathway of apoptosis as induced by TNFa. For this purpose, we will establish culture systems in which some TNFa sensitive cell lines will be infected with adenovirus mutants having specific or no deletions in the E3 region. We will determine the effect of adenoviral infection and TNFa, alone or in combination, on cell death and ceraccumulation. Also, the effects of adenoviral infection on ceramide-induced apoptosis will be determined by using exogenous ceramide treatment. The role of the E3 region of adenovirus will then be determined by comparing the effects of wild type virus with those of a mutant virus lacking the E3 region. G. Dbeibo. Ceramide inhibits interleukin-2 induction: mechanisms of action

Interleukin-2 (IL-2) provides a signal necessary for T-cell proliferation and, consequently, immune response activation. Expression of the IL-2 gene is regulated at a transcriptional level by multiple trans-acting factors binding to the promoter region of the gene including. Glycosphingolipids have been shown to be immunosuppressants by inhibiting T-cell proliferation. In this project, we examine the inhibitory role of ceramide, a sphingolipid breakdown product, on IL-2 induction. Preliminary results show that ceramide inhibits IL-2 production by Jurkat leukemic T-cells. The mechanism of this inhibition appears to be through the prevention of NF-kB activation that is necessary for IL-2 gene transcription. This activity of ceramide is specific to protein kinase C mediated NF-kB activation and is not evident in tumor necrosis factor a-mediated NF-kB activation. The effect of experimental status epilepticus on ceramide levels in the developing brain

Kainic Acid (KA) induced Status Epilepticus (SE) has been shown to cause apoptosis in the adult rat brain. Immature rats younger than P30, however, are resistant to the effects of SE and do not manifest neuronal death like mature adult animals do. The mechanisms underlying those differences are not well understood. The Sphingolipid ceramide has been demonstrated to participate in the regulation of cell response to stress and to drive the apoptotic response in many nonneuronal and some neuronal cell culture models. The potential role of ceramide in producing neuronal injury post SE has been studied. The goal of this study is, thus, to investigate whether SE is followed by increases in ceramide levels and whether there are differences in ceramide levels after SE in different developmental stages. Potential role of ceramide in status epilepticus related brain injury

We have recently demonstrated that Kainic Acid (KA) induced Status Epilepticus (SE) is associated with increases in levels of the sphingolipid ceramide in adult and prepubescent rats. The goal of this study is, thus, to investigate whether the demonstrated increases in ceramide are related to SE associated neuronal injury. We hope to accomplish this by measuring levels of ceramide and by performing concomitant apoptosis stains in two different models of KA induced SE that allow, depending on the experimental conditions, the tested animals to undergo SE with or without subsequent cell death and apoptosis. In addition, we plan to perform a third experiment in which the synthesis of ceramide is inhibited in animals undergoing KA induced SE in order to find out whether this can prevent the occurrence of apoptosis. The effect of chronic hypoxia on the developing brain

Although the effects of acute hupoxia on the developing brain have been shown to be age dependent, the effects of chronic hypoxia are much less understood. Chronic hypoxia represents a common problem in patients with cyanotic congenital heart disease, but it is not known yet what its consequences really are and whether those are preventable. We, thus, plan to investigate the hypotheses that chronic hypoxia starting at birth: (1) results in long term impairment in memory and learning (as assessed by the Morris Water Maze test), (2) causes long term behavioral abnormalities (as assessed by the Handling and Open Field test), (3) causes long term epilepsy, increased seizure susceptibility and neuronal injury (as assessed by video-tape technique, fluorthyl test, and histologic technique), and (4) worsens consequences of superimposed acute hypoxic episodes. The eventual goal of this research is to understand the mechanisms of brain damage that occur in cyanotic congenital heart disease, and to subsequently, if appropriate, investigate the mechanisms and means of prevention of those impairments possibly by the long term use of neuroprotective agents. Hyperoxia-induced apoptosis in the neonatal rat lung

The study aims at characterizing the role of apoptosis in hyperoxia-induced cell death in the neonate. Neonatal rats are put in a hyperoxic environment up to 7 days. Lung sections are obtained at multiple time points from hyperoxia-exposed and control animals. Different parameters are being studied: morphologic changes of apoptosis through TUNEL assay and DNA gel electrophoresis and production of ceramide, a mediator of apoptosis and release of BCL2 and BAX, two modulators of apoptosis.
