Title of the project: The effect of septic shock on the absorption of aminoacids in the rat jejunum A
new model of septic shock and resuscitation is developed. Shock is induced
by injecting the rat with bacteria (E. coli) and bacterial endotoxin
via a femoral vein catheter while monitoring the blood pressure via a femoral
artery catheter. The heart rate is monitored via two electrodes attached
to the skin. The shock state is defined by maintaining the blood pressure
at 60% of the control value for one hour. Resuscitation is accomplished
by infusing saline solution until the blood pressure is restored to the
control value. Pharmacologic manipulation is achieved by intravenous injection
of calcium channel blockers, nitric oxide inhibitors and stimulants, oxygen
free radical scavengers and other agents. Aminoacid absorption (alanine
and glutamine) is measured using the single pass perfutechnique for in
vivo studies and the two compartment system with isolated intestinal
strips for the in vitro studies. These studies may help us understand
the mechanisms of amino acid absorption in the rat jejunum in states of
bacteremia, septic shock and post shock syndrome. This may have therapeutic
value through pharmacologic manipulation of the gut during these pathologic
